ABSTRACT

ObjectiveTo observe the effects of Youguiwan on bone metabolism and bone morphogenetic protein-2 (BMP-2)/Smad signaling pathway in ovaries-removed rats with osteoporosis and study the mechanism of Youguiwan in the prevention and treatment of osteoporosis. MethodA postmenopausal rat model of osteoporosis was prepared by bilateral ovariectomy. The 40 female SD rats were randomly divided into five groups, including sham operation group, model group, alendronate sodium group (0.1 mg·kg-1), and high-dose and low-dose (5.36 and 2.68 g·kg-1) groups of Youguiwan. The drug was given seven days after modeling, once a day for 12 weeks. After the treatment, the changes in femur tissue structure were observed by micro-CT, including bone mineral density (BMD), bone volume/total volume (BV/TV), trabecular number (Tb.N), trabecular thickness (Tb.Th), bone surface/bone volume (BS/BV), and trabecular separation (Tb.Sp). Ossification was observed by saffrane-solid green staining, and serum levels of bone metabolism markers, including bone alkaline phosphatase (BALP), osteocalcin (BGP), type Ⅰ procollagen amino terminal propeptide (PINP), and tartrate-resistant acid phosphatase 5b (TRACP-5b), were determined by enzyme-linked immunosorbent assay (ELISA). The protein and mRNA expression levels of Runx2, BMP-2, and Smad1 in rat femur were detected by Western blot and Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). ResultCompared with the sham operation group, bone trabecula in the model group was sparse. BMD, BV/TV, Tb.N, and Tb.Th were decreased (P<0.05, P<0.01). BS/BV (P<0.05) and Tb.Sp were increased. The content of BGP, BALP, PINP, and TRACP-5b in serum was significantly increased (P<0.01). The mRNA and protein expressions of Runx2, BMP-2, and Smad1 in rat femur were significantly decreased (P<0.05, P<0.01). Compared with the model group, the number of bone trabeculae in the high-dose and low-dose groups of Youguiwan was increased, and the bone microstructure was improved. BMD, BV/TV, Tb.N, and Tb.Th were increased significantly (P<0.05, P<0.01), and BS/BV and Tb.Sp were increased. The content of bone metabolic markers decreased (P<0.05, P<0.01). ConclusionYouguiwan has certain preventive and therapeutic effects on postmenopausal osteoporosis, and its mechanism may be related to promoting bone formation by regulating the BMP-2/Smad signaling pathway.

ABSTRACT

Objetive: To investigate the mechanism of antidepressant effect of verbascoside based on high-throughput sequencing technology (RNA-Seq),and to explore the possible targets and signaling pathways. MethodsForty C57BL/6 mice were randomly divided into control group, model group, fluoxetine group and verbascoside group,10 mice in each group. Except for the control group, all the other three groups were constructed with chronic unpredictable mild stimulation (CUMS) combined with solitary feeding for four weeks. Control group, fluoxetine group and verbascoside group were administered by gavage once daily for three weeks during the second week of modeling. The mice were assessed by sugar-water preference test, forced swimming test, open field test, elevated cross maze test, and water maze test. Enzyme linked immunosorbent assay(ELISA) was performed to detect the levels of major neurotransmitters and inflammatory factors in mice serum, and mRNA high-throughput sequencing was performed in the nucleus accumben and colon to screen differentially expressed genes and perform pathway enrichment analysis. Real-time polymerase chain reaction(Real-time PCR) was used to detect the mRNA expression of Gad,Slc32a1(VGAT) and brain-derived neurotrophic factor (BDNF) in nucleus accumbens. ResultCompared with the control group, the anxiety and depression-like behaviors in the model group increased, while the learning and memory ability decreased significantly. The content of neurotransmitter in serum decreased significantly. The content of pro-inflammatory factors increased significantly. The mRNA expression of Gad and Slc32a1(VGAT) in nucleus accumbens decreased significantly, while that of BDNF increased significantly. Compared with the model group, the anxiety and depression-like behavior of mice in verbascoside group was significantly relieved. The neurotransmitter content increased significantly, and the pro-inflammatory factors decreased significantly. The mRNA expression of Gad and Slc32a1(VGAT) in nucleus accumbens increased significantly, while that of BDNF decreased significantly.A total of 48 differentially expressed genes in nucleus accumbens and 43 differentially expressed genes in colon were screened by high-throughput sequencing. Differential genes in nucleus accumbens mainly focus on neuroactive ligand-receptor interaction, γ-aminobutyric acid(GABA)ergic synapse, synaptic vesicle cycle and other pathways. Colonic differential genes are mainly concentrated in GABAergic synapses, synaptic vesicle circulation, cyclic adenosine monophosphate(cAMP) signal pathway and other signal pathways. Compared with the control group, the mRNA expression of Gad and Slc32a1(VGAT) in nucleus accumbens of model group decreased significantly, while the mRNA expression of BDNF increased significantly. ConclusionVerbascoside has significant antidepressant effects. Its antidepressant effect may be related to the increase of monoamine neurotransmitters, the decrease of pro-inflammatory factors and the restoration of neurotransmitter homeostasis by increasing GABA,and it mainly acts through the signaling pathways such as neuroactive ligand-receptor interaction,GABAergic synapses, synaptic vesicle cycle and cAMP signaling pathway.

ABSTRACT

Objective To study the association between vitamin D status and bone mineral density(BMD) in aged people. Methods A total of 118 patients admitted in cadre department aged (77.7±11.2) years were selected from July 2010 to May 2011.The BMD at lumbar spine (L1-4) and femoral neck was measured by dual energy X-ray absorptiometry (DEXA).According to BMD,patients were divided into two groups:osteoporosis and non-osteoporosis group. The data of serum 25-hydroxy vitamin D3 [25(OH)D3]concentration,height,weight and age of patients were collected at the same time. Results The age and body mass index (BMI) in osteoporosis group were (81.6 ±5.6) years and (22.5 ± 4.0 ) kg/m2,while (79.4 ± 6.9 ) years and (24.1± 4.2 ) kg/m2 in nonosteoporosis group (t=1.80 and -2.01,P>0.05).The concentrations of serum 25(OH)D3 in two groups were(21.6± 10.3)nmol/L and (32.0± 13.8) nmol/L,respectively(t=-4.20,P<0.01).And there were 95.3 % (41/43 )and 81.3 % (61/75) of patients whose serum 25 (OH)D3 level were ≤50nmol/L in osteoporosis and non- osteoporosis group,respectively(x2 =4.58,P<0.05).Furthermore,the 25 (OH) D3 level was positively correlated with BMD at femoral neck(r=0.22,P<0.05),but not correlated with BMD at L1-4 ( r=0.18,P>0.05).Conclusions Vitamin D status is correlated with BMD at femoral neck in aged people.
